Allergen uptake into pDC is an active process. PDC were isolated from buffy coats by BDCA-4 positive selection. Lin-1−/BDCA-2+ staining revealed a pDC purity of > 95%. After overnight incubation at 4 °C or 37 °C in AIM-V medium containing 10 ng/ml IL-3 and 10 µg/ml Alexa Fluor 555 labeled allergens (Api, OVA or Der p 1) allergen uptake was analyzed by flow cytometry.
